Cancer cells are in constant communication and modulate TME through several mechanisms, including transfer of tumour-promoting cargos through extracellular vesicles (EVs) or oncogenic signal detection by primary cilia. Spheresomes are a specific EV that arise from rough endoplasmic reticulum–Golgi vesicles. They accumulate beneath cell membrane and are released to the extracellular medium through multivesicular spheres. This study describes spheresomes in low-grade gliomas using electron microscopy. We found that spheresomes are more frequent than exosomes in these tumours and can cross the blood–brain barrier. Moreover, the distinct biogenesis processes of these EVs result in unique cargo profiles, suggesting different functional roles. We also identified primary cilia in these tumours. These findings collectively contribute to our understanding of glioma progression and metastasis. 000127802 536__ $$9info:eu-repo/grantAgreement/ES/DGA/LMP248_21 000127802 540__ $$9info:eu-repo/semantics/openAccess$$aby$$uhttp://creativecommons.org/licenses/by/3.0/es/ 000127802 590__ $$a3.8$$b2023 000127802 592__ $$a0.9$$b2023 000127802 591__ $$aMULTIDISCIPLINARY SCIENCES$$b25 / 134 = 0.187$$c2023$$dQ1$$eT1 000127802 593__ $$aMultidisciplinary$$c2023$$dQ1 000127802 655_4 $$ainfo:eu-repo/semantics/article$$vinfo:eu-repo/semantics/publishedVersion 000127802 700__ $$aIruzubieta, Pablo 000127802 700__ $$0(orcid)0000-0001-7453-2470$$aCastiella, Tomás 000127802 700__ $$0(orcid)0000-0002-2787-9671$$aMonzón, Marta$$uUniversidad de Zaragoza 000127802 700__ $$0(orcid)0000-0002-7453-1766$$aMonleón, Eva$$uUniversidad de Zaragoza 000127802 700__ $$aBerga, Carmen 000127802 700__ $$aSchuhmacher, Alberto J. 000127802 700__ $$0(orcid)0000-0002-9951-1075$$aJunquera, Concepción$$uUniversidad de Zaragoza 000127802 7102_ $$11003$$2443$$aUniversidad de Zaragoza$$bDpto.
