Papel de las bombas de efluvios en la resistencia a los antibióticos y bactericidas en Pseudomonas syringae

Abstract: Since antibiotics discovery, their widespread use, has resulted in resistant pathogens accordingly multi-drug resistant strains has appeared. Bacterial pathogens mainly use drug efflux as a mechanism for resistance against antimicrobial compounds. Blocking this efflux can increase the intracellular concentrations of the available antibiotics resulting in an enhancement of their efficiency. For this reason, the discovery and development of efflux pump inhibitors (EPIs) is an interesting approach to be studied.
The purpose of this study is to find an effective treatment against bacterial resistance of Pseudomonas syringae. The method used is MIC determination by measuring absorbance in 96 well plates at different concentration of the following compounds: CuSO4, TPP+, tetracycline, chloramphenicol and streptomycin. The EPIs used are phenylalanine arginyl β-naphthylamide (PaβN) and chlorpromazine (CPZ). The strains that have been worked with are two laboratory strains: P. syringae pathovar phaseolicola (ATCC® 21781™ known as HB10Y) and P. syringae pv. atrofaciens (P. Syr. Pv.); and four field isolates: TL1, SWG11, WWG2 and WWG74. The first part of the experiment consists of MIC determination of mentioned compounds in lack of EPIs. Afterwards, the MIC values are calculated again but in presence of EPIs.
Finally, the variation of MIC values in both cases is compared. The use of EPIs increases the effectiveness of the antibiotics with which it is combined because their presence reduces their MICs value. That happens in the case of TPP+ and tetracycline in HB10Y, and TPP+ in TL1. In many other cases the bacterial growth is reduced in EPIs presence, but MIC values are not affected. Finally, in the case of SWG11 or WWG2 strains, EPI presence has had no impact on antibiotics MIC values. The conclusions drawn are that for an effective treatment a previous study of the strains, antibiotics, concentrations and EPI is needed.
