SARS-CoV-2, the causative agent of COVID-19, induces both specific T- and B-cell responses, being antibodies against the virus detected a few days after infection. Passive immunization with hyperimmune plasma from convalescent patients has been proposed as a potentially useful treatment for COVID-19. Using an in-house quantitative ELISA test, we found that plasma from 177 convalescent donors contained IgG antibodies specific to the spike receptor-binding domain (RBD) of SARS-CoV-2, although at very different concentrations which correlated with previous disease severity and gender. Anti-RBD IgG plasma concentrations significantly correlated with the plasma viral neutralizing activity (VN) against SARS-CoV-2 in vitro. Similar results were found using an independent cohort of serum from 168 convalescent health workers. These results validate an in-house RBD IgG ELISA test in a large cohort of COVID-19 convalescent patients and indicate that plasma from all convalescent donors does not contain a high enough amount of anti-SARS-CoV-2-RBD neutralizing IgG to prevent SARS-CoV-2 infection in vitro. The use of quantitative anti-RBD IgG detection systems might help to predict the efficacy of the passive immunization using plasma from patients recovered from SARS-CoV-2. 000101576 536__ $$9info:eu-repo/grantAgreement/ES/MINECO/PID2019-105451GB-I00$$9info:eu-repo/grantAgreement/ES/MINECO/CTQ2013-44367-C2-2-P$$9info:eu-repo/grantAgreement/ES/MINECO/BFU2016-75633-P$$9info:eu-repo/grantAgreement/ES/MCIU-AEI/SAF2017-83120-C2-1-R$$9info:eu-repo/grantAgreement/ES/ISCIII/COV20-00308$$9info:eu-repo/grantAgreement/ES/DGA/LMP58-18$$9info:eu-repo/grantAgreement/ES/DGA-FEDER/E34-R17$$9info:eu-repo/grantAgreement/ES/DGA-FEDER/B29-17R 000101576 540__ $$9info:eu-repo/semantics/openAccess$$aby$$uhttp://creativecommons.org/licenses/by/3.0/es/ 000101576 590__ $$a5.168$$b2021 000101576 592__ $$a0.903$$b2021 000101576 594__ $$a2.8$$b2021 000101576 591__ $$aBIOLOGY$$b21 / 94 = 0.223$$c2021$$dQ1$$eT1 000101576 593__ $$aBiochemistry, Genetics and Molecular Biology (miscellaneous)$$c2021$$dQ1 000101576 593__ $$aAgricultural and Biological Sciences (miscellaneous)$$c2021$$dQ1 000101576 655_4 $$ainfo:eu-repo/semantics/article$$vinfo:eu-repo/semantics/publishedVersion 000101576 700__ $$aUranga-Murillo, Iratxe 000101576 700__ $$aArias, Maykel 000101576 700__ $$0(orcid)0000-0002-5838-0857$$aGonzález-Ramírez, Andrés M.$$uUniversidad de Zaragoza 000101576 700__ $$0(orcid)0000-0001-6815-6720$$aMacías-León, Javier$$uUniversidad de Zaragoza 000101576 700__ $$0(orcid)0000-0002-0182-201X$$aMoreo, Eduardo$$uUniversidad de Zaragoza 000101576 700__ $$aRedrado, Sergio 000101576 700__ $$0(orcid)0000-0003-3791-2997$$aGarcía-García, Ana$$uUniversidad de Zaragoza 000101576 700__ $$0(orcid)0000-0001-9224-5854$$aTaleb, Víctor$$uUniversidad de Zaragoza 000101576 700__ $$aLira-Navarrete, Erandi$$uUniversidad de Zaragoza 000101576 700__ $$0(orcid)0000-0002-3122-9401$$aHurtado-Guerrero, Ramón 000101576 700__ $$0(orcid)0000-0001-7897-9173$$aAguilo, Nacho$$uUniversidad de Zaragoza 000101576 700__ $$adel Mar Encabo-Berzosa, Maria 000101576 700__ $$aHidalgo, Sandra 000101576 700__ $$aGalvez, Eva M. 000101576 700__ $$aRamirez-Labrada, Ariel 000101576 700__ $$ade Miguel, Diego 000101576 700__ $$0(orcid)0000-0001-5134-1006$$aBenito, Rafael$$uUniversidad de Zaragoza 000101576 700__ $$aMiranda, Patricia 000101576 700__ $$0(orcid)0000-0002-2557-4890$$aFernández, Antonio$$uUniversidad de Zaragoza 000101576 700__ $$aDomingo, José M. 000101576 700__ $$aSerrano, Laura 000101576 700__ $$aYuste, Cristina 000101576 700__ $$0(orcid)0000-0001-6209-4282$$aVillanueva-Saz, Sergio$$uUniversidad de Zaragoza 000101576 700__ $$0(orcid)0000-0002-9600-8116$$aPaño-Pardo, José R.$$uUniversidad de Zaragoza 000101576 700__ $$0(orcid)0000-0003-0154-0730$$aPardo, Julián$$uUniversidad de Zaragoza 000101576 7102_ $$11011$$2630$$aUniversidad de Zaragoza$$bDpto.
